I have some question which I can't tell the difference. Maybe some PROs here can answer me. Thank you!
1. ONE SINGLE INSERT SITUATION..
If I put one plugin on the fifth slot and leave all the other 4 slots empty, will that sound differently if I move this plugin from fifth slot to the first slot?
2. MULTIPLE INSERT SITUATION..
If I put EQ in first slot, Reverb in the second slot. Will that sound differently if I reverse them?
thank you for reading my question!

There'll be no differences whether you assign the plug in on the first slot or the others.

It depends on what kind of results you want. Same thing happens like Compressors and the EQ plugins, it's all up to your demand for a " before or After" assignment. And sometimes you need to use, for instances, two L1 next to each other, and before or after that, for an "on your face kind of vocal", an EQ boost the Hi frequency for a better and cleaner image and tone...

PS :
Not only in Protools, same as Digital Performer , Cubase , Sonar ...
All the logic and rule of INSERT , Basicly is all the same..
The Digitalize Mixer is base on the concept of Analog mixer .
Insert is just like AUX , Send , Return .
The output assignment is like Group , bus .... ( or whatever you call )
Maybe a little bit different . but the result is same.
The only difference is Digital mixer is virture.
you can save the money to get real cable ...
the decrease the loss of sound quality through Cable ......
